Choosing the Right Media for Your Research

Consistent and reproducible cell cultures are critical to experimental success. That's why Cook MyoSite's Custom Cell Solutions (CCS) MyoTonic™ and CryoTonic™ family of media products are designed to support researchers through every stage of the primary human skeletal muscle cell culture workflow, from expansion and differentiation to cryopreservation. Cook MyoSite's media formulations were developed using decades of experience in primary human muscle-related technologies and are optimized for skeletal muscle-derived cell culture applications.

Rather than asking which medium is best, a better question is: What are you trying to accomplish with your cells?

If You Need to Expand Cells and Freeze Them for Later Use

For routine cell growth, expansion, and cryopreservation, CCS recommends:

    • MyoTonic™ Basal Medium (MB-2222)
    • MyoTonic™ Growth Supplement (MS-3333)
    • CryoTonic™ CryoStorage Medium (CR-9999)

This combination supports cell expansion while providing a solution for storing valuable cell populations for future experiments.

If Your Goal Is Myotube Differentiation

Differentiation is a two-stage process in which cells are first expanded in growth medium and then transitioned to differentiation medium. For this workflow, CCS recommends:

    • MyoTonic™ Basal Medium (MB-2222)
    • MyoTonic™ Growth Supplement (MS-3333)
    • MyoTonic™ Differentiation Medium (MD-5555)

Differentiated cultures are characterized by the formation of elongated, multinucleated myotubes.

If You Want to Use Your Own Serum

Some researchers require greater control over serum components or may encounter restrictions related to serum sourcing. For these applications, CCS offers:

    • MyoTonic™ Basal Medium (MB-2222)
    • MyoTonic™ Serum-Free Growth Supplement (MS-8888)
    • MyoTonic™ Serum-Free Differentiation Medium (MD-9999) when differentiation is required

These products allow researchers to incorporate their own fetal bovine serum (FBS) while maintaining a standardized growth and differentiation system.

If You Need Control Over Insulin Levels

For studies where insulin concentration is an important experimental variable, CCS offers:

    • MyoTonic™ Insulin-Free Basal Medium (ML-6666)
    • MyoTonic™ Serum-Free Growth Supplement (MS-8888)

This option provides researchers with the flexibility to culture cells under insulin-free conditions or introduce insulin according to their experimental design.
